Nearly 27,000 people in Tewkesbury voted against the closure of Alderman Knight school in a referendum organised by the town council, and only 304 were in favour.
Head teacher Ieuan Walsh hoped the vote would convince the area's schools organisation committee that Gloucestershire County Council was acting against the wishes of the community.
If the committee turns down the county council's plan, the final decision would rest with an adjudicator appointed by the Department for Education and Skills.
Gloucestershire council said the schools organisation committee would consider the result of the referendum along with "all other objections and evidence".
