The company will form part of an expert group dedicated to developing the new course, which will be on offer to pupils from 2011.
Michael Boyd, artistic director at the RSC said the humanities diploma would introduce young people to themes from Shakespeare's plays, including democracy, dictatorship and humanity.
He said: "We look forward to working with other employers to ensure the diploma is an excellent basis for progression to employment in a wide range of fields."
Children's Secretary Ed Balls claimed the existing five diplomas are proving popular with employers, teachers and pupils.
He said: "It is great news that the RSC are on board to develop the humanities diploma, I'm sure they will help make it a sought after qualification."
